The Pasteur-IRCAN Molecular and Cellular Imaging Core Facility : PICMI

The Pasteur-IRCAN Molecular and Cellular Imaging Core Facility (PICMI) gathers the scientific and technical expertise and state of the art devices required for scientific projects using imaging techniques.

We provide services ranging from simply accessing our facilities to performing your imaging experiments. You can thus choose between :

-       Becoming an autonomous user thanks to training sessions adapted to your experimental needs and a follow up on the apparatus used. You can then use it according to your needs and manage your schedule.

-       Having our platform operators perform your sample preparation and image acquisitions to gain time and benefit from their expertise.

Equipment

PICMI Equipment

* Inverted Confocal Microscope ZEISS LSM Exciter

This microscope allows to visualize proteins and small molecules at the cellular level using fluorescent labeling.

The specificity of a confocal system is to perform thin optical sections of the sample thanks to a small pinhole aperture placed in front of the photomultiplier. The signal obtained is transformed in numerical images than can then be processed and quantified.

This allows to improve axial resolution and to get rid of the fluorescence originating from regions outside of the observed focal plane. Then, upon acquisition of a stack of images, the observed objects can be rendered in 3D with limited image post-processing.

* ZEISS High-throughput Live Epi-fluorescence Microscope

It allows the automated acquisition of large numbers of wide field pictures of multiple regions of interest and at multiple time points (Time Lapse and Point Visiting). Phase contract and epi-fluorescence pictures can also be associated in this setting.

* ZEISS Time Lapse Epi-fluorescence Microscope

This system can produce wide field pictures in time series and data can be analyzed using the MetaMorph® software.

- A-Plan 5x/0,12 PHo, A-Plan x10/0,25 PH1, Plan-Neofluar x20, x40, Fluar x40, Plan Apochromate x63 (oil-immersion) objectives
- Motorized stage
- Environment control for CO2 and temperature
- Acquisition and analysis using MetaMorph®

* ZEISS Epi-fluorescence Cytogenetics Microscope

This cytogenetics station allows to acquire wide field epi-fluorescence images and to perform semi-automated cytogenetic analyzes of metaphase spreads (karyotyping, mFISH) using the Isis and Ikaros softwares (MetaSystems).

- Plan-Apochromat  x10/0,45 PH1, Plan- Neofluar  x20/0,5 PH2, Plan-Neofluar x40/0,75 PH2, x40, Fluar x40, Plan- Apochromat x63 and 100x (oil-immersion) obectives.

Booking

PICMI Booking

Every new user must first contact one of the core facility operators in order to be advised on the system that fits best his/her needs.

Autonomous access to the facility and the booking schedule is granted only if the user follows a compulsory training session and signs the PICMI user agreement.
