Résumé


Les Télomères

Les télomères sont des structures nucléoprotéiques dont la fonction essentielle est de former une structure protégeant l’extrémité des chromosomes contre les phénomènes de dégradation, de fusions inter- ou intra-chromosomiques et de recombinaison, pouvant être à l’origine d’instabilité génétique. Dans la plupart des cancers humains, la structure et la fonction des télomères subissent des modifications qui facilitent la prolifération des cellules cancéreuses, leur survie, et leur adaptation à différents microenvironnements cellulaires. Les télomères constituent à ce titre une cible intéressante pour le développement de drogues anti-cancer. Notamment, les protéines constituant la base de la structure nucléoprotéique du télomère (appelé le télosome ou le complexe « Shelterin ») apparaissent comme des cibles de choix pour le développement de nouveaux agents anticancéreux.


Les objectifs principaux de ce projet sont de mieux comprendre le fonctionnement des télomères et d'identifier et de caractériser les rôles des télomères et de la télomérase dans la transformation maligne des cellules humaines. Nous centrerons nos approches sur l'étude de la chromatine télomérique, de l'intégration des fonctions télomériques au niveau cellulaire et du rôle joué par l'instabilité télomérique dans l'oncogenèse. Nous proposons d'étudier le fonctionnement des télomères et de la télomérase, d'explorer les modifications de la chromatine télomérique dans les tumeurs et des manipulations à façon de ces modifications dans des systèmes de tumorigenèse expérimentale.


Notre insistance à comprendre les conséquences moléculaires du dysfonctionnement du télosome devrait nous conduire à la compréhension de nouveaux mécanismes impliqués dans l’oncogénèse et à la découverte de nouveaux champs d’investigation pour le traitement des cancers. À travers l’exploration du rôle du télosome dans l’oncogenèse, nous espérons identifier des biomarqueurs et générer de nouveaux agents thérapeutiques.

RF2 condenses telomeric DNA Atomic force microscopy images of TRF2 complexes with a 650 pb telomeric duplex DNA.

These complexes were shown to correspond to dimers, tetramers, hexamers and octamers of TRF2. Several molecules are shown in panels a and d. In panel a free DNA is also shown and in panel d, only the central molecule corresponds to the octameric form of the complex.

Note the way DNA shortens with the increasing mass of the complex indicating DNA wrapping. Models of the complexes are drawn above the images.
